Abstract Pancreatic ductal adenocarcinoma, characterised by a suppressive tumour microenvironment (TME), is associated with dismal survival rates and limited treatment options. Our proposal is to leverage CRISPR-Cas9 technology to disrupt the immunosuppressive TME through gene knockout of associated protein. This novel strategy can improve efficacy when combined with conventional treatments, as well as allowing for a more targeted and adaptable model to traditional inhibitory drug development.
